5-Hydroxytryptophan (5-HTP) is a hydroxylated derivative of L-tryptophan and an aromatic amino acid containing an indole ring substituted with a hydroxyl group. Its chemical structure is closely related to L-tryptophan, with hydroxylation occurring at the 5-position of the indole system. In biological systems, 5-HTP can be formed through enzymatic hydroxylation of L-tryptophan, a reaction catalyzed by tryptophan hydroxylases or engineered aromatic amino acid hydroxylases. This transformation has been extensively investigated in biochemical and metabolic-engineering studies aimed at developing microbial production systems for hydroxylated tryptophan derivatives. The distinctive spectroscopic properties of the 5-hydroxyindole group have also made 5-HTP useful in studies of proteins and molecular interactions. Researchers have incorporated 5-HTP into proteins as a noncanonical amino acid, enabling spectroscopic analysis and selective chemical modification of protein structures.

5-Hydroxytryptophan is used in biochemical and molecular research as an intrinsic spectroscopic probe. Its characteristic absorption and fluorescence properties have been exploited to investigate protein structure, protein–DNA interactions and molecular association processes.
It has also been incorporated into proteins as a noncanonical amino acid in engineered bacterial systems. Studies have demonstrated genetically controlled incorporation of 5-HTP into proteins and subsequent site-selective modification of the incorporated residue with aromatic diazonium compounds.
5-HTP has additionally been investigated in surface-chemistry and materials research, including studies of its adsorption on silver nanoparticles and its incorporation into epoxy-based systems evaluated for corrosion-related properties.
