What role do acids play in organic reactions?

Acids are used when a reaction requires increased substrate reactivity, improved leaving-group ability, stabilization of an intermediate or shifting of equilibrium toward the desired product. They may protonate oxygen, nitrogen or sulfur atoms, activate multiple bonds, increase the electrophilicity of a carbonyl carbon or form more reactive complexes with the substrate. This enables transformations that would otherwise be slow or poorly selective under neutral conditions.

How do Brønsted acids differ from Lewis acids?

Brønsted acids operate by proton transfer, so their role is especially visible in protonation, solvolysis, hydrolysis, esterification, dehydration and acetal formation. Lewis acids do not need to contain a proton, because they act by accepting an electron pair from the substrate. In practice, this means coordination to electron-rich atoms such as oxygen, nitrogen, sulfur or halogens, which can increase the electrophilicity of nearby reaction centers.

Mineral and organic acids in synthesis

Common Brønsted acids include mineral acids such as hydrochloric, sulfuric, phosphoric and nitric acid, as well as organic acids such as acetic, formic, trifluoroacetic, methanesulfonic and p-toluenesulfonic acid. They differ in acid strength, volatility, counterion nucleophilicity, water content, solvent compatibility and effect on substrate stability. Therefore, acid selection depends not only on strength but also on whether the reaction environment should be aqueous, anhydrous, polar, mild or strongly activating.

Lewis acids as substrate activators

Lewis acids such as aluminum chloride, boron trifluoride, tin(IV) chloride, zinc chloride, iron(III) chloride, metal triflates and selected metal complexes are used to activate substrates containing donor centers. They are especially important in reactions involving carbonyl compounds, epoxides, acetals, acyl halides, alkenes and aromatic compounds. Lewis acid coordination may change reaction rate, regioselectivity, stereoselectivity and the susceptibility of a substrate to nucleophilic attack.

Which reactions commonly use acids?

Acids are widely used in esterification, transesterification, ester and acetal hydrolysis, acetalization, alcohol dehydration, addition to alkenes, epoxide opening, Friedel-Crafts reactions, rearrangements, cyclizations and condensations. In many of these transformations, the acid is not merely a pH-adjusting additive but directly affects formation of a reactive intermediate or activation of a functional group.

Acids in carbonyl chemistry

Carbonyl compounds are particularly responsive to acid activation because the oxygen atom can be protonated or coordinated by a Lewis acid. This activation increases the electrophilicity of the carbonyl carbon and facilitates reaction with a nucleophile. This mechanism is important in acetalization, Mukaiyama-type aldol reactions, selected condensations and reactions with alcohols, amines and organosilicon compounds.

When are solid and heterogeneous acids used?

Solid acids such as zeolites, ion-exchange resins, metal oxides, heteropoly acids, aluminosilicates and acids supported on carriers are used when easier catalyst separation from the reaction mixture is beneficial. They may reduce problems associated with corrosiveness, recovery and neutralization of classical mineral acids. Their effectiveness depends on acid-site accessibility, surface area, porosity, substrate diffusion and resistance to catalyst deactivation.

Chiral acids and enantioselective synthesis

Chiral Brønsted acids and chiral Lewis acids are used in asymmetric synthesis, where the goal is not only to accelerate the reaction but also to control the three-dimensional structure of the product. Such systems include chiral phosphoric acids, selected carboxylic acids, metal complexes with chiral ligands and catalysts combining acid activation with hydrogen-bonding interactions. Their behavior depends on substrate fit within the catalytic environment and stabilization of the transition state.

How should an acid be selected for a planned reaction?

The choice of acid depends on substrate type, required acid strength, sensitive functional groups, solvent, water content, temperature and expected mechanism. A strong acid may activate a substrate efficiently, but it can also cause decomposition, excessive protonation, polymerization, rearrangement or side reactions. A milder acid can be more useful when selective activation of one functional group is required without disturbing other parts of the molecule.

Limitations of acids in organic synthesis

Acids may influence a reaction in difficult-to-predict ways when the substrate contains several basic or nucleophilic sites. Competitive protonation, changes in solubility, salt formation, substrate degradation, isomerization, polymerization or loss of selectivity may occur. In systems using Lewis acids, the presence of water and donor solvents is also important because they may deactivate the catalyst through coordination or hydrolysis.

Safety and limitations of use

Acids used in organic synthesis may be corrosive, volatile, toxic, strongly oxidizing, hygroscopic, fuming or moisture-sensitive, depending on the specific substance. Mineral acids, sulfonic acids, superacids and strong Lewis acids require particular control of working conditions, material compatibility and contact with water, bases or reducing substances. Each acid should be selected and used according to its safety data sheet and the requirements of the specific reaction.
