Terpenes follow the “isoprene rule,” arising from head-to-tail coupling of C₅ building blocks to form monoterpenes (C₁₀), sesquiterpenes (C₁₅), diterpenes (C₂₀), triterpenes (C₃₀), and tetraterpenes (C₄₀). In biosynthesis, these units originate from isopentenyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP), generated via the mevalonate or MEP/DOXP pathways. Terpene synthases catalyze cationic cyclization and rearrangement cascades, producing immense structural diversity (acyclic, monocyclic, polycyclic frameworks). While “terpenes” strictly denote hydrocarbons, oxygenated derivatives are termed terpenoids (e.g., alcohols, aldehydes, ketones, epoxides). Their reactivity is dominated by alkene chemistry—electrophilic additions, oxidations, and rearrangements—often influenced by conformational constraints in polycyclic systems. Terpenes contribute to membrane structure (sterols), photosynthesis (carotenoids), signaling (phytohormones), and ecological interactions, and they are widely used in perfumery, flavorings, and as starting materials for pharmaceuticals and polymers.
