NMR ANALYSIS 

Nuclear Magnetic Resonance (NMR) spectroscopy is a powerful analytical technique used to determine the structure, dynamics, and interactions of molecules. It is based on the principle that certain atomic nuclei, when placed in a strong magnetic field, absorb and re-emit electromagnetic radiation at specific frequencies. These frequencies are characteristic of the chemical environment of the nuclei, making NMR a valuable tool for elucidating molecular structures.

Preparation of a Sample for NMR Analysis

If you have chosen to have the sample prepared by us  you can skip this information, sample can be sent in any form and container as long as it is prepared safely.

Proper sample preparation is essential for obtaining accurate and high-quality NMR spectra. The steps involved in preparing a sample are as follows:

  1. Selection of Solvent:
    Use a deuterated solvent (e.g., CDCl3, DMSO-d6, D2O) to dissolve the sample. Deuterated solvents are necessary because they do not produce interfering signals in the 1HNMR spectrum, and they provide a lock signal for the spectrometer.

  2. Sample Dissolution:
    Dissolve approximately 5-20 mg of the compound in 0.6-1 mL of the deuterated solvent. Ensure the compound is completely dissolved to avoid peak broadening caused by undissolved particles. If necessary, warm the solution gently or use ultrasonication to aid dissolution. 13C NMR requires higher concentrations, typically around 20 mg of sample per 1 mL of solvent. Please provide us with the concentrations you are using.

  1. Filtration (if required):
    If the sample contains insoluble impurities or particles, filter the solution through a small syringe filter or centrifuge it to obtain a clear solution. This step helps prevent artifacts in the spectrum.

  2. Transfer to an NMR Tube:
    Using a clean pipette, transfer the solution into a high-quality NMR tube (typically 5 mm in diameter). Ensure the tube is clean and free of scratches to avoid signal distortions.

  3. Adjusting Sample Volume:
    The recommended volume of solution in the NMR tube is approximately 0.6-1 mL to ensure the sample fills the active region of the NMR coil. Avoid overfilling or underfilling the tube.

  4. Adding Internal Standards (if required):
    If quantitative analysis or chemical shift referencing is needed, add a small amount of an internal standard (e.g., TMS – tetramethylsilane for δ=0\delta = 0δ=0 ppm reference) to the solution.

  5. Sealing the Tube:
    Cap the NMR tube securely to prevent solvent evaporation, which could affect the concentration and chemical shift.

  6. Inspect the Sample:
    Check the solution for air bubbles, as they can distort the magnetic field. Tap the tube gently to remove any bubbles.

Once the sample is prepared, it is ready to be placed in the NMR spectrometer for analysis. Proper preparation minimizes noise and artifacts, ensuring clear and interpretable spectra.
